Landevejscyklingens linjeløb for kvinder under sommer-OL 2016 fandt sted den 7. august 2016.

Ruten var 136,9 km lang, og startede ved Forte de Copacabana, og gik derefter vestover langs kysten via Ipanema og Barra da Tijuca. Derefter startede to 27,4 km lange runder (Circuito Grumari) med to stigninger (Grumari og Grota Funda), og to kilometer med brosten. Så vendte ruten tilbage østover forbi Barra da Tijuca. Derefter var der en runde på 25,5 km i Circuito Vista Chinesa med to stigninger. På toppen var der under 15 km til mål på Copacabana.
