Landevejscykling for mænd i cykling under Sommer-OL 2012 fandt sted den 28. juli 2012. Løbet var 250 km lang og startet ved The Mall før den gik syd-vest over for at krydse Themsen ved Putney Bridge. Derefter gik det gennem Richmond Park, Bushy Park og forbi Hampton Court Palace før ni runder over Box Hill. Etter Box Hill vendte rytterne tilbage til opløbet på The Mall.

Mark Cavendish var den største forhåndsfavorit. Med støtte fra et stærkt britisk hold bestående af fire etapevindere (syv etaper) fra Tour de France 2012, inkluderet nummer 1 og 2 sammenlagt.

Da omgangene på Box Hill var over bestod førergruppen af 26 ryttere, som havde et minuts forspring. Storbritannien havde brugt kræfterne og fik ikke hjælp fra andre hold og fik derved ikke indhentet udbruddet. Aleksandr Vinokurov og Rigoberto Urán kørte fra udbruddet få kilometer før mål, og Vinokurov vandt spurten.

Jakob Fuglsang blev bedste dansker med en tolvte plads.
